Figure 1. Na and Cl, and K+ excretion by rs3857080 genotype. The figure depicts the 24 h excretion of Na and Cl after furosemide intake (A) and of potassium on moderate sodium chloride restriction (B) in relation to the genotype of rs3857080. Sodium and chloride excretion were evaluated as a combined end point. Data are mean/standard error of mean. Symbol sizes are proportional to the group sizes. Number of subject in the genotypic groups are GG: 72; GA: 19; AA: 1 for Na and Cl after furosemide and GG: 76; GA: 23; AA: 0 for potassium on moderate sodium chloride restriction. Concerning this polymorphism (rs3857080), we observed 87, 22 and 1 carriers of the GG, GA and AA genotypes in the first study and 93, 25 and 1 carriers of GG, GA and AA in the second study (the AA carrier in the second study did not complete any study day), respectively.

Figure 2. Differences in urinary sodium and chloride, and potassium excretion with rs3857080 and Ile180Val (rs5522). The figure depicts the differences in the 24 h excretion of sodium chloride (Na and Cl) and of potassium (K+ ) associated with the two polymorphisms rs3857080 and Ile180Val (rs5522). Data are mean differences (± standard error of the mean). They were calculated by four times seven covariance analyses. Predictors included in the model were the two NR3C2 polymorphisms, urinary drug excretion and the eight polymorphisms formerly published to affect urinary excretion in our studies [ 5 , 22 , 25 , 26 ]. Numbers give the differences associated with two minor alleles (A or valine alleles) compared with two major alleles (G or isoleucine alleles). HCT: Hydrochlorothiazide; NaCl: Sodium chloride.

Figure 3. Time course of potassium and sodium and chloride excretion by rs3857080 genotypes. The figure depicts the electrolyte excretion over 24 h separately for homozygous G allele carriers of rs3857080 (green diamonds) and heterozygous G/A carriers (black diamonds). The figure depicts the time courses of sodium plus chloride excretion after furosemide in (A) and the time course of K+ excretion after 100 mg HCT in (B) , after triamterene in (C) , and after moderate sodium chloride restriction in (D) ....
